If you want to drive in expert mode (which I recommend) try the following:
Press P to raise the pantograph. You'll see the needle on the right rise.
Press O to close the circuit breaker. You'll see the red breaker light on the left go out.
Press and hold M to release the train brakes. Wait for the air needles on the left to stop rising.
Press Z to move the reverser to forward. You'll see the lever on the right rear move towards you.
Press E to move the traction lever (right front) one notch towards you. You should see the traction coil light on the left go out. If you see VI light on the left come on you've gone one notch too far. Use shift E to move it back.
Press and hold the space bar. This is the deadman's handle.
Press Q to increase power. You should see the green indicator move leftwards.
Press D to reduce power.
The ' key (this will probably vary with different keyboard layouts) should apply the train airbrakes.
If you hear a bell go off then release the space bar then re-press it (vigilance device).
There's more to it, such as dynamic brakes and a really good speed control mode, but hopefully this will get you moving.

You probably haven't made up the consist correctly. Most connections are with articulated bogies. Make sure there are no missing or overlapping bogies between cars.

I think the key layouts are a bit strange because the author has designed it for a non qwerty keyboard layout.
