Is your train completely off the switch?

Yep, the end of the loco or car has to be several feet from the turnout points, or the lever will reverse itself to its original position or not activate at all.

Also, the 'hot spot' that the mouse cursor uses to activate a turnout can be a bit flaky in the 3D map, if it isn't working use 'G' for the turnout closest to the front of the loco or 'Shift+G' for the one nearest its rear.

If it is an automatic switch, it won't unless you are in a freeroam. Even then, you must be a bit away from the switch.

You can also go to the 2D layout where each switch (automatic and manual) is displayed as a dot.
You can click on that dot while holding shift and the switch will change.
