I found this while searching around the web today. It's a really nice guide on the signaling used on US railroads:

This should definitely prove useful for folks having trouble interpreting the signals on US routes in Railworks.

While playing the game, I'm always confused by signals (can't remember the meaning), so I just barrel through anything that's not all red.
